The Increased Phosphoration of Akt by Fenofibrate and its Effect on Radiation Sensitivity in Brain Tumor Cells

Abstract
Purpose: Fenofibrate (FF) is a PPAR¥á ligand and used as a hypolipidemics. We found that FF increases phosphyrylated Akt (pAkt) in a glioblastoma multiforme cell line (U87). pAkt has been known to increase cell survival and decrease radiation sensitivity. Therefore, we investigated whether increased pAkt by FF is associated with radiation sensitivity.

Materials and Methods: A glioblastoma cell line, U87 cells were treated with FF alone or in combination with a pAkt inhibitor, LY294002 (LY) and the change of pAkt was analyzed with Western blot. The radiation sensitivity was measured by using clonogenic assay.

Results: pAkt was increased by 30 ¥ìM of FF treatment for 48 h and it was decreased by the addition of 10 ¥ìM of LY. Surviving fraction (SF) by 2 Gy was increased from 0.38 to 0.44 by preincubation with FF before irradiation. The increased SF by FF was decreased by the addition of LY; 0.25.

Conclusion: pAkt is an important factor which determines radiation sensitivity. FF decreases radiation sensitivity by increasing pAkt.
